Midkine (MDK) is a heparin-binding growth factor that is highly expressed in many malignant tumors, including lung cancers. MDK activates the PI3K pathway and induces anti-apoptotic activity, in turn enhancing the survival of tumors. Therefore, the inhibition of MDK is considered a potential strategy for cancer therapy. In the present study, we demonstrate a novel small molecule compound (iMDK) that targets MDK. iMDK inhibited the cell growth of MDK-positive H441 lung adenocarcinoma cells that harbor an oncogenic KRAS mutation and H520 squamous cell lung cancer cells, both of which are types of untreatable lung cancer. However, iMDK did not reduce the cell viability of MDK-negative A549 lung adenocarcinoma cells or normal human lung fibroblast (NHLF) cells indicating its specificity. iMDK suppressed the endogenous expression of MDK but not that of other growth factors such as PTN or VEGF. iMDK suppressed the growth of H441 cells by inhibiting the PI3K pathway and inducing apoptosis. Systemic administration of iMDK significantly inhibited tumor growth in a xenograft mouse model in vivo. Inhibition of MDK with iMDK provides a potential therapeutic approach for the treatment of lung cancers that are driven by MDK.  相似文献

Recent evidence indicates that melanoma comprises distinct types of tumors and suggests that specific morphological features may help predict its clinical behavior. Using a SNP‐array approach, we quantified chromosomal copy number alterations (CNA) across the whole genome in 41 primary melanomas and found a high degree of heterogeneity in their genomic asset. Association analysis correlating the number and relative length of CNA with clinical, morphological, and dermoscopic attributes of melanoma revealed that features of aggressiveness were strongly linked to the overall amount of genomic damage. Furthermore, we observed that melanoma progression and survival were mainly affected by a low number of large chromosome losses and a high number of small gains. We identified the alterations most frequently associated with aggressive melanoma, and by integrating our data with publicly available gene expression profiles, we identified five genes which expression was found to be necessary for melanoma cells proliferation. In conclusion, this work provides new evidence that the phenotypic heterogeneity of melanoma reflects a parallel genetic diversity and lays the basis to define novel strategies for a more precise prognostic stratification of patients.  相似文献

Anatomic aortic anomalies are seen in many medical conditions and are known to cause disturbances in blood flow. Turner syndrome (TS) is a genetic disorder occurring only in females where cardiovascular anomalies, particularly of the aorta, are frequently encountered. In this study, numerical simulations are applied to investigate the flow characteristics in four TS patient- related aortic arches (a normal geometry, dilatation, coarctation and elongation of the transverse aorta). The Quemada viscosity model was applied to account for the non-Newtonian behavior of blood. The blood is treated as a mixture consisting of water and red blood cells (RBC) where the RBCs are modeled as a convected scalar. The results show clear geometry effects where the flow structures and RBC distribution are significantly different between the aortas. Transitional flow is observed as a jet is formed due to a constriction in the descending aorta for the coarctation case. RBC dilution is found to vary between the aortas, influencing the WSS. Moreover, the local variations in RBC volume fraction may induce large viscosity variations, stressing the importance of accounting for the non-Newtonian effects.  相似文献

Introduction

Tissue plasminogen activator (tPA)-activity and plasminogen activator inhibitor type 1 (PAI-1) antigen are considered to be haemostasis-related markers of endothelial activation and relate to presence of cerebral white matter hyperintensities (WMH) as was earlier shown in a cross-sectional study. We investigated whether tPA-activity and PAI-1 levels are associated with WMH progression in a longitudinal study.

Methods

In 127 first-ever lacunar stroke patients in whom baseline brain MRI and plasma levels of tPA-activity and PAI-1-antigen were available, we obtained a 2-year follow-up MRI. We assessed WMH progression by a visual WMH change scale. We determined the relationship between levels of tPA-activity and PAI-1 and WMH progression, by logistic regression analysis.

Results

Plasma tPA-activity was associated with periventricular WMH progression (OR 2.36, 95% CI 1.01–5.49, with correction for age and sex and baseline presence of WMH), but not with deep or any (periventricular and/or deep) WMH progression. PAI-1 levels were lower in patients with WMH progression, but these results were not significant.

Conclusion

We found a relationship between plasma tPA-activity and progression of periventricular WMH. More research is needed to determine whether there is a (direct) role of tPA in the development and progression of WMH.  相似文献

The 2 histone deacetylase inhibitors (HDACIs) approved for the treatment of cutaneous T-cell lymphoma (CTCL) including mycosis fungoides/sezary syndrome (MF/SS), suberoylanilide hydroxamic acid (SAHA) and romidepsin, are associated with low rates of overall response and high rates of adverse effects. Data regarding combination treatments with HDACIs is sparse. Butyroyloxymethyl diethylphosphate (AN-7) is a novel HDACI, which was found to have selective anticancer activity in several cell lines and animal models. The aim of this study was to compare the anticancer effects of AN-7 and SAHA, either alone or combined with doxorubicin, on MF/SS cell lines and peripheral blood lymphocytes (PBL) from patients with Sezary syndrome (SPBL). MyLa cells, Hut78 cells, SPBL, and PBL from healthy normal individuals (NPBL) were exposed to the test drugs, and the findings were analyzed by a viability assay, an apoptosis assay, and Western blot. AN-7 was more selectively toxic to MyLa cells, Hut78 cells, and SPBL (relative to NPBL) than SAHA and also acted more rapidly. Both drugs induced apoptosis in MF/SS cell lines, SAHA had a greater effect on MyLa cell line, while AN-7 induced greater apoptosis in SPBL; both caused an accumulation of acetylated histone H3, but AN-7 was associated with earlier kinetics; and both caused a downregulation of the HDAC1 protein in MF/SS cell lines. AN-7 acted synergistically with doxorubicin in both MF/SS cell lines and SPBL, and antagonistically with doxorubicin in NPBL. By contrast, SAHA acted antagonistically with doxorubicin on MF/SS cell lines, SPBL, and NPBL, leaving <50% viable cells. In conclusion, AN-7 holds promise as a therapeutic agent in MF/SS and has several advantages over SAHA. Our data provide a rationale for combining AN-7, but not SAHA, with doxorubicin to induce the cell death in MF/SS.  相似文献

Gene duplications are an important factor in plant evolution, and lineage-specific expanded (LSE) genes are of particular interest. Receptor-like kinases expanded massively in land plants, and leucine-rich repeat receptor-like kinases (LRR-RLK) constitute the largest receptor-like kinases family. Based on the phylogeny of 7,554 LRR-RLK genes from 31 fully sequenced flowering plant genomes, the complex evolutionary dynamics of this family was characterized in depth. We studied the involvement of selection during the expansion of this family among angiosperms. LRR-RLK subgroups harbor extremely contrasting rates of duplication, retention, or loss, and LSE copies are predominantly found in subgroups involved in environmental interactions. Expansion rates also differ significantly depending on the time when rounds of expansion or loss occurred on the angiosperm phylogenetic tree. Finally, using a dN/dS-based test in a phylogenetic framework, we searched for selection footprints on LSE and single-copy LRR-RLK genes. Selective constraint appeared to be globally relaxed at LSE genes, and codons under positive selection were detected in 50% of them. Moreover, the leucine-rich repeat domains, and specifically four amino acids in them, were found to be the main targets of positive selection. Here, we provide an extensive overview of the expansion and evolution of this very large gene family.Receptor-like kinases (RLKs) constitute one of the largest gene families in plants and expanded massively in land plants (Embryophyta; Lehti-Shiu et al., 2009, 2012). For plant RLK gene families, the functions of most members are often not known (especially in recently expanded families), but some described functions include innate immunity (Albert et al., 2010), pathogen response (Dodds and Rathjen, 2010), abiotic stress (Yang et al., 2010), development (De Smet et al., 2009), and sometimes multiple functions (Lehti-Shiu et al., 2012). The RLKs usually consist of three domains: an N-terminal extracellular domain, a transmembrane domain, and a C-terminal kinase domain (KD). In plants, the KD usually has a Ser/Thr specificity (Shiu and Bleecker, 2001), but Tyr-specific RLKs were also described (e.g. BRASSINOSTEROID INSENSITIVE1; Oh et al., 2009). Interestingly, it was estimated that approximately 20% of RLKs contain a catalytically inactive KD (e.g. STRUBBELIG and CORYNE; Chevalier et al., 2005; Castells and Casacuberta, 2007; Gish and Clark, 2011). In Arabidopsis (Arabidopsis thaliana), 44 RLK subgroups (SGs) were defined by inferring the phylogenetic relationships between the KDs (Shiu and Bleecker, 2001). Interestingly, different SGs show different duplication/retention rates (Lehti-Shiu et al., 2009). Specifically, RLKs involved in stress responses show a high number of tandemly duplicated genes whereas those involved in development do not (Shiu et al., 2004), which suggests that some RLK genes are important for the responses of land plants to a changing environment (Lehti-Shiu et al., 2012). There seem to be relatively few RLK pseudogenes compared with other large gene families, and copy retention was argued to be driven by both drift and selection (Zou et al., 2009; Lehti-Shiu et al., 2012). As most SGs are relatively old and RLK subfamilies expanded independently in several plant lineages, duplicate retention cannot be explained by drift alone, and natural selection is expected to be an important driving factor in RLK gene family retention (Lehti-Shiu et al., 2009).Leucine-rich repeat-receptor-like kinases (LRR-RLKs), which contain up to 30 leucine-rich repeat (LRRs) in their extracellular domain, constitute the largest RLK family (Shiu and Bleecker, 2001). Based on the KD, 15 LRR-RLK SGs have been established in Arabidopsis (Shiu et al., 2004; Lehti-Shiu et al., 2009). So far, two major functions have been attributed to them: defense against pathogens and development (Tang et al., 2010b). LRR-RLKs involved in defense are predominantly found in lineage-specific expanded (LSE) gene clusters, whereas LRR-RLKs involved in development are mostly found in nonexpanded groups (Tang et al., 2010b). It was also discovered that the LRR domains are significantly less conserved than the remaining domains of the LRR-RLK genes (Tang et al., 2010b). In addition, a study of four plant genomes (Arabidopsis, grape [Vitis vinifera], poplar [Populus trichocarpa], and rice [Oryza sativa]) showed that LRR-RLK genes from LSE gene clusters show significantly more indications of positive selection or relaxed constraint than LRR-RLKs from nonexpanded groups (Tang et al., 2010b).The genomes of flowering plants (angiosperms) have been shown to be highly dynamic compared with most other groups of land plants (Leitch and Leitch, 2012). This dynamic is mostly caused by the frequent multiplication of genetic material, followed by a complex pattern of differential losses (i.e. the fragmentation process) and chromosomal rearrangements (Langham et al., 2004; Leitch and Leitch, 2012). Most angiosperm genomes sequenced so far show evidence for at least one whole-genome multiplication event during their evolution (Jaillon et al., 2007; D’Hont et al., 2012; Tomato Genome Consortium, 2012). At a smaller scale, tandem and segmental duplications are also very common in angiosperms (Arabidopsis Genome Initiative, 2000; International Rice Genome Sequencing Project, 2005; Rizzon et al., 2006). Although the most common fate of duplicated genes is to be progressively lost, in some cases they can be retained in the genome, and adaptive as well as nonadaptive scenarios have been discussed to play a role in this preservation process (for review, see Moore and Purugganan, 2005; Hahn, 2009; Innan, 2009; Innan and Kondrashov, 2010). Whole-genome sequences also revealed that the same gene may undergo several rounds of duplication and retention. These LSE genes were shown to evolve under positive selection more frequently than single-copy genes in angiosperms (Fischer et al., 2014). That study analyzed general trends over whole genomes. Here, we ask if, and to what extent, this trend is observable at LRR-RLK genes. As this gene family is very dynamic and large, and in accordance with the results of Tang et al. (2010b), we expect the effect of positive selection to be even more pronounced than in the whole-genome average. We analyzed 33 Embryophyta genomes to investigate the evolutionary history of the LRR-RLK gene family in a phylogenetic framework. Twenty LRR-RLK SGs were identified, and from this data set, we deciphered the evolutionary dynamics of this family within angiosperms. The expansion/reduction rates were contrasted between SGs and species as well as in ancestral branches of the angiosperm phylogeny. We then focused on genes whose number increased dramatically in an SG- and/or species-specific manner (i.e. LSE genes). Those genes are likely to be involved in species-specific cellular processes or adaptive interactions and were used as a template to infer the potential occurrence of positive selection. This led to the identification of sites at which positive selection likely acted. We discuss our results in the light of angiosperm genome evolution and current knowledge of LRR-RLK functions. Positive selection footprints identified in LSE genes highlight the importance of combining evolutionary analysis and functional knowledge to guide further investigations.  相似文献

Last-century climate change has led to variable increases of the intrinsic water-use efficiency (Wi; the ratio of net CO2 assimilation to stomatal conductance for water vapor) of trees and C3 grassland ecosystems, but the causes of the variability are not well understood. Here, we address putative drivers underlying variable Wi responses in a wide range of grassland communities. Wi was estimated from carbon isotope discrimination in archived herbage samples from 16 contrasting fertilizer treatments in the Park Grass Experiment, Rothamsted, England, for the 1915 to 1929 and 1995 to 2009 periods. Changes in Wi were analyzed in relation to nitrogen input, soil pH, species richness, and functional group composition. Treatments included liming as well as phosphorus and potassium additions with or without ammonium or nitrate fertilizer applications at three levels. Wi increased between 11% and 25% (P < 0.001) in the different treatments between the two periods. None of the fertilizers had a direct effect on the change of Wi (ΔWi). However, soil pH (P < 0.05), species richness (P < 0.01), and percentage grass content (P < 0.01) were significantly related to ΔWi. Grass-dominated, species-poor plots on acidic soils showed the largest ΔWi (+14.7 μmol mol−1). The ΔWi response of these acidic plots was probably related to drought effects resulting from aluminum toxicity on root growth. Our results from the Park Grass Experiment show that Wi in grassland communities consistently increased over a wide range of nutrient inputs, soil pH, and plant community compositions during the last century. The intrinsic water-use efficiency (Wi) of plants is controlled by photosynthetic carbon assimilation and stomatal conductance via the leaf-level coupling of CO2 and water fluxes. A general, but variable, increase of Wi under rising atmospheric CO2 has been observed in long-term studies (Peñuelas et al., 2011; Franks et al., 2013; Saurer et al., 2014), but little is known about other environmental or ecosystem factors, which may interact with the effect of increasing CO2 on Wi. An improved understanding of putative interactive mechanisms is important because changes in Wi may have significant effects on the global terrestrial carbon and water cycles (Gedney et al., 2006; Betts et al., 2007). This study explores the interactive effects of the increase in atmospheric CO2 (observed over the last century), nutrient loading, and soil pH together with other related effects on plant species richness and functional group composition on the coupling of plant CO2 and water fluxes in a seminatural grassland in southeastern England.Wi is a leaf-level efficiency that has also been termed potential water-use efficiency or physiological water-use efficiency, as it excludes the direct influence of vapor pressure deficit (VPD), a parameter determined by environmental conditions, on leaf-level water-use efficiency (Farquhar et al., 1989; Franks et al., 2013). Wi reports the relationship between net CO2 assimilation rate (An) and stomatal conductance for water vapor (gH2O):(1)According to the first law of Fick, An can be given as the product of the stomatal conductance for CO2 (gCO2) and the concentration gradient between the atmosphere (ca) and the leaf internal gas space (ci): An= gCO2 (caci). Using gCO2 (caci) instead of An in Equation 1, replacement of gH2O/gCO2 by the numerical value of gH2O/gCO2 (1.6) and rearrangement yields the following alternative expression of Wi:(2)Equation 2 reveals that past changes of Wi must have been controlled by two parameters: the change of ca and the concurrent change of 1 – ci/ca, the relative gradient for CO2 diffusion into the leaf (Franks et al., 2013). A change in the relative gradient is determined by the changes in An relative to gH2O, as leaves respond to changing ca and other environmental factors. In particular, Equation 2 shows that any variation in the climate change response of Wi is determined by the ci/ca response, if the comparison is made for vegetation at the same location and in the same period of time.Studies with C3 vegetation, including trees/forests and C3 grasslands, have revealed a general increase of Wi in the last century (Bert et al., 1997; Duquesnay et al., 1998; Feng, 1999; Arneth et al., 2002; Saurer et al., 2004; Barbosa et al., 2010; Köhler et al., 2010; Andreu-Hayles et al., 2011). In many cases, ci/ca, estimated by 13C discrimination (Farquhar et al., 1989), varied relatively little. Indeed, it has been suggested, based on theoretical grounds and empirical evidence from studies over geological/evolutionary to short time scales, that adaptive feedback responses will tend to maintain ci/ca approximately constant (Ehleringer and Cerling, 1995; Franks et al., 2013), as plants optimize carbon gain with respect to water loss (Cowan and Farquhar, 1977). Yet, ci/ca-dependent variation in the Wi response to climate change has also been noted (Peñuelas et al., 2011; Köhler et al., 2012) over the last century, indicating that additional factors, perhaps including other global change drivers, can modify the Wi response over this time scale, at least transiently. A meta-analysis by Peñuelas et al. (2011) reports ci/ca-dependent increases of Wi for different forests between 6% and 36% from the early 1960s to 2000s. A recent study by Saurer et al. (2014) on European forest trees found increases in Wi ranging from 1% to 53% during the last century. The strongest increase of Wi was recorded in regions where summer soil-water availability decreased in the last century. For different grassland communities, the ci/ca-dependent increases of Wi varied between 13% and 28% at one site (Köhler et al., 2012) from 1915 to 2009. Evidently, such variation can have important repercussions for the coupling of terrestrial CO2 and water fluxes. Yet, little is known about the mechanism(s) underlying the variation. At the Park Grass Experiment (PGE) at Rothamsted, England, Köhler et al. (2012) observed a nitrogen supply-dependent enhancement of the Wi response on plots receiving nitrate fertilizer and maintained at a near-neutral soil pH by liming. However, the actual relationship between nitrogen supply and Wi response did not hold when the unlimed control (soil pH approximately 5.2) was included in the comparison. Remarkably, however, there was a significant positive relationship between the grass content of the community and the Wi response of the experimental plots in the investigation. These results suggested that the effect of nutrient supply on the Wi response of the grassland communities was indirect, perhaps working via effects on soil pH and/or vegetation composition (plant species richness or functional group composition). The PGE provides a unique opportunity to study century-scale variation in the ci/ca-dependent variation of Wi for a wide range of diverse grassland communities. Much of the extant ecosystem-scale variability of plant species richness and soil pH in temperate grasslands of Europe (Ceulemans et al., 2014) is included in the range of plot-scale plant species richness and soil pH at the PGE (which is reported in this investigation). The different long-term applications of fertilizer and lime over the past century have resulted in substantial changes in soil pH, species richness, and grass content on the experimental plots, but in most cases, within-plot changes over the study period considered here (1915–2009) were comparatively small (Crawley et al., 2005; Silvertown et al., 2006). All experimental plots are located at the same site and are exposed to the same weather conditions. Consequently, trends in climate as a direct driver for differences in Wi between plots can be ruled out. Here, we explore putative mechanisms underlying eventual ci/ca-dependent variation of Wi during the last century at the PGE by, first, quantifying the sustained effect of a wide range of contrasting fertilizer treatments (n = 16) on the change of Wi during the last century and, second, analyzing the relationships between the observed Wi response of treatments and the respective nutrient status, soil pH, plant species richness, and plant functional group composition of the grassland communities.  相似文献
